With regards to potency the conformation of a drug plays a role, where lower energy conformations are preferred to give adequate affinity. Likewise, there is a need to make specific intermolecular interactions with the target macromolecule. By understanding both these concepts we can understand how drugs work and it helps us to design and develop better medicines.
